A method for increasing the formation of HDL in a subject comprising administering to the subject an agent that inhibits the PEST-mediated proteolysis of ABCA1 protein in the subject's cells is described. An article of manufacture comprises a packaging and a pharmaceutical agent. The methods can be used for determining whether a protease inhibitor increases the rate of cholesterol efflux from a cell and for increasing ABCA1-mediated cholesterol efflux from a cell and for increasing HDL formation in a subject, thereby treating atherosclerosis in a subject.
